Gathering and Washing Soil 10/1/18

Rationale: After having another plaque assay turn up negative I decided to go out and collect soil again. However, I didn’t have means to return to Cameron Park today, so I decided to collect a sample on campus.

Procedure:

  1. Collected a sample from a large chinquapin oak near Truett Seminary then

    returned to the lab and placed 2mL of the sample into a tube.

  2. Filled tube with 10mL of LB broth then shook for 15 minutes.
  3. Filled another tube with equal mass water then spun together at 3000g for 5

    minutes.

  4. Filtered using 22nm filter and a syringe into a 50mL tube.
  5. Added .5mL arthrobacter and placed into shaking incubator for 48 hours.

Observations:
The tree was 13.79 meters tall with a circumference at 137cm of 185.3cm. The canopy diameter was 8.625 meters. The tree seemed decently healthy, with a few damaged leaves.

Interpretations and Next Steps: Next lab I will collect metadata and make a plaque assay in hopes of getting a plaque. I’m currently in a loop of collecting soil and conducting plaque assays until I get a plaque.
